دراسة التوافر الحيوي لمركب الدوكسي سايكلين في جرعاته المختلفة  عن طريق الفم لدى دجاج اللحم

الملخص

هدفت هذه الدراسة إلى تقييم التوافر الحيوي والحركية الدوائية للدوكسي سايكلين المُعطى فموياً بجرعات 20 ملغ/كغ و 40 ملغ/كغ متدرجة في دجاج اللحم. استُخدمت في هذه الدراسة ثلاثة مجموعات من طيور دجاج اللحم، تضم كل مجموعة (25) طيراً بعمر (28يوم) وهي كالآتي:

المجموعة الأولى (G1): وهي مجموعة الشاهد السلبي والتي تضم 25طيراً وهذه المجموعة لم تأخذ أي علاج (مجموعة غير معالجة).

المجموعة الثانية (G2): تضم هذه المجموعة 25طيراً حيثُ تم إعطاؤها جرعة علاجية واحدة فقط من المضاد الحيوي الدوكسي سايكلين بالتجريع عن طريق الفم مقدارها 20 ملغ/كغ (مجموعة معالجة بجرعة علاجية واحدة).

المجموعة الثالثة (G3): تضم هذه المجموعة 25طيراً حيثُ تم إعطاؤها جرعة واحدة فقط ضعف علاجية من المضاد الحيوي الدوكسي سايكلين بالتجريع عن طريق الفم مقدارها 40 ملغ/كغ (مجموعة معالجة بجرعة ضعف علاجية واحدة).

جُمعت عينات الدم من الوريد الجناحي بعد إجراء التعقيم المناسب بعد (15دقيقة، نصف ساعة، 1ساعة، 2ساعة، 3ساعة، 4ساعة، 5ساعة، 6ساعة، 7ساعة، 8ساعة، 9ساعة، 10ساعة، 12ساعة، 16ساعة، 24ساعة،32 ساعة)، ووضعت في أنابيب غير حاوية على مانع تخثر، وتم تثفيل العينات بسرعة 3000 دورة في الدقيقة ولمدة 10 دقائق للحصول على المصل، وحُللت تراكيز الدواء بتقنية HPLC، كما تم قياس نشاط أنزيم ALT الكبدي بعد 24 ساعة.

أظهرت النتائج وجود فُروق معنوية (P < 0.05) بين جميع المجموعات والأزمنة، مع تسجيل الشاهد تراكيز صفرية تؤكد دقة الطريقة. بلغ تركيز الدواء ذروته (C_max) بعد ساعتين من تجريع المضاد الحيوي ضمن مجموعات التجربة. أما بالنسبة لنشاط الإنزيم الكبديALT  فقد لوحظ وجود ارتفاع معنوي ضمن مجموعات التجربة الثانية والثالثة اللتان تلقتا جرعة علاجية20  ملغ/كغ وضعف علاجية 40 ملغ/كغ مقارنةً مع مجموعة الشاهد.
