دراسة الإصابة بـ الليشمانية المدارية Leishmania tropica في القداد السوري الذهبي وفئران BALB/c

الملخص

أُجريت هذه الدراسة لتقييم التغيرات السريرية والمرضية الناتجة عن الخمج التجريبي بطفيلي الليشمانية المدارية في القداد السوري الذهبي وفئران BALB/c، ومقارنة قابلية كل منهما للإصابة كنموذج حيواني. كما هدفت إلى تتبع توزع الطفيلي في العقدة اللمفية المأبضية والطحال ونقي العظم باستخدام تقنية تفاعل البوليمراز المتسلسل المعشّش Nested-PCR.

شملت الدراسة مجموعتين، ضمت كل منهما 15 أنثى بعمر ثمانية أسابيع، إحداهما من القداد السوري الذهبي والأخرى من فئرانBALB/c.

جرى إكثار الطفيلي مخبرياً، ثم حُقنت الحيوانات في وسادة القدم اليمنى، وتمت متابعتها سريرياً لمدة أربعة أشهر، مع القتل الرحيم لعدد من الحيوانات على فترات شهرية لإجراء الفحوص الجزيئية على الأعضاء المستهدفة.

أظهرت النتائج استمرار الكشف عن الحمض النووي لطفيلي الليشمانية المدارية في القداد السوري الذهبي خلال الأشهر الثلاثة الأولى بعد الخمج، في الأعضاء الثلاثة المستهدفة بالدراسة، وهي العقدة اللمفية المأبضية والطحال ونقي العظم، في المقابل، اقتصر الكشف عن الحمض النووي للطفيلي في فئران BALB/cعلى الشهر الأول فقط بعد الخمج، وذلك في العقدة اللمفية المأبضية والطحال فقط. كما ظهر تورم في وسادة القدم وآفات جلدية عابرة لدى بعض حيوانات القداد، إضافة إلى تضخم الطحال في عدد منها، بينما لم تسجل تغيرات سريرية أو مرضية واضحة في فئرانBALB/c.

تشير النتائج إلى وجود اختلاف في قابلية الإصابة ومسار الخمج بين النموذجين الحيوانيين، إذ أظهر القداد السوري الذهبي استمراراً أطول للحمل الطفيلي مقارنةً بفئرانBALB/c. وتبرز أهمية اختيار النموذج الحيواني المناسب لدراسة عدوى الليشمانية المدارية، ولاسيما في أبحاث الإمراضية والاستجابة المناعية وتقييم العلاجات واللقاحات المستقبلية.
